On-Chip Bioassay in 3D Microchannel Networks

An on-chip whole-cell bioassay method has been developed using a mutagen-responsive bioluminsecent E. coli strain. Strains are immobilized in an assembly in which an Si chip is placed between two PDMS chips. In the assembly, microchannels on the two PDMS layers are connected via perforated microwells on the Si chip to form a 3D microfluidic network. This on-chip format enables high-throughput bioassay for multiple samples using multiple tester strains in a single chip assembly. The format is applicable to various types of chemical and biochemical analysis.
